It’s in the southern end of the square, there should be a sign and some steps down, looks like it’s open noon-4:30. When I was there in 2019 there were a couple of rooms full of cats being sheltered for various reasons and you could go hang out with them, dunno about now.
There’s also a cat shelter in the Non-catholic Cemetery (where Keats, Shelley, and Gramsci are buried) by the Pyramid of Cestius but that one wasn’t open to the public last I saw. Usually cats wandering in the cemetery tho.
I’m not sure if it is a shelter or just a feral cat colony. But there sure are a lot of cats there.
